Measurement significance:
FAS is a key enzyme in fatty acid synthesis, catalysing the production of long-chain fatty acids from acetyl coenzyme A and malonyl coenzyme A. FAS is commonly expressed in a variety of tissues and cells, and is abundantly expressed in mammalian liver, kidney, brain, lungs and mammary glands, as well as adipose tissue.
Principle of Assay:
FAS catalyses the production of long-chain fatty acids and NADP+ from acetyl CoA, malonyl CoA and NADPH; NADPH has an absorption peak at 340 nm, whereas NADP+ does not; the rate of decrease in light absorption at 340 nm was measured to calculate FAS activity.
