Significance of measurement:
SSS (EC 2.4.1.21) is usually present in the free state in the plastid matrix, catalyses the elongation of starch chains and is mainly responsible for the synthesis of branched starch.
Principle of Assay:
SSS catalyses the reaction of ADPG with the starch primer (dextran), transferring glucose molecules to the starch primer and generating ADP at the same time. Pyruvate kinase, hexokinase and glucose 6-phosphate dehydrogenase added to the reaction system catalyse the reduction of NADP+ to NADPH sequentially, and the amount of NADPH generated is directly proportional to the amount of ADP generated in the previous step of the reaction, which is measured at 340 nm. SSS activity can be calculated by measuring the increase in NADPH at 340 nm.
